Position Summary:
This position will be laboratory-based, working directly with a team of scientists within the Functional Area Support Team (FAST) within Platform Biology. The individual will gain first-hand experience in Proteolysis-targeting chimera (PROTAC) drug discovery by helping to execute and analyze biochemical and cellular plate-based pharmacology experiments. PROTACs have been developed as a drug modality for targeted protein degradation. A bifunctional PROTAC molecule consists of a ligand (typically small-molecule inhibitor) of the protein of interest (POI) and a covalently linked ligand of an E3 ubiquitin ligase (E3). Within a fast-paced, collaborative environment the individual will be expected to learn quickly in order to meaningfully contribute to the progression of potential disease area therapeutics.
This position will be located at our headquarters in New Haven, CT.
Principal Responsibilities:
Key responsibilities of this role include, but are not limited to:
- Training on various assays and data analysis. Assays include 1) biochemical ligand displacement formats such as the Time-resolved Fluorescence Resonance Energy Transfer (TR-FRET) assay that works on the principle that when suitable pairs of fluorophores are in close proximity of one another, excitation of a lanthanide donor fluorophore results in energy transfer to an acceptor fluorophore; 2) cell-based HiBiT (Promega) assays in which a POI endogenous to a cell line of interest is tagged with a luciferase-derived protein using CRISPR technology that allows for sensitive bioluminescent detection of protein degradation.
- Test compounds in plate-based pharmacology experiments where the concentration of the test compound is varied and response in the assay is measured. The analyzed dose-response effect data is used to inform medicinal chemists.
- Learn to analyze and communicate experimental data to team members.
- Write up experimental procedures for colleagues and provide thorough documentation using an electronic lab notebook.

Qualifications:

- Basic understanding of cell and molecular biology and/or biochemistry
- Prior experience with cell culture highly desired, but not a pre-requisite
- Capable of independent work and communicating progress or problems
- Excellent data handling and recording skills as well as oral and written communication skills.
- Proficiency with Microsoft Office Suite
Education:
- Preference to a Biochemistry/Chemistry or Cell and Molecular Biology Junior/Senior student in an undergraduate program
- Completed advanced coursework in cell biology and/or biochemistry.
